You start off with the ability to send out two commanders on different missions. As you unlock more levels in your City, you will gain the ability to send out more commanders at the same time.
You can start to send out three commanders around level 12-14 - Sorry I can't remember exactly, and a fourth at level 17 of your City Hall.
