What building should I upgrade for the defense of my city? How does it work?

The tower. Unfortunately a key ingredient (alloy barrels) will require diamonds.

The Garrison. It has a function for City Defense. You have to add it 80 units every 30 minutes till you get to 8,300 for maximum protection. And yes, also the Towers, but the drawback there is needing the diamonds to purchase Alloy Barrels.

This is to repair damage after an attack. you do need to maintain it as you will be relocated if it reaches 0.

How do you add troops to the garrison? What do you mean by units?

The new Garrison hall building in the 'Strategy' section of the Build menu now adds resistance bonuses to your defending troops.
