It means you need to defeat 500 tier 3 or above units from other players. Kill or attack 500 troops above tier 3

Does that mean attack bases?

Yes, or potentially gathering troops too but attacking bases is better.

It means the level of your troops, you get higher level troops if you upgrade the training Centre. Tier 1 is the first level of troops and it gets higher if you upgrade.
