All the buildings require connection by road to be accessible and usable. (decorations don't need road though) Some buildings you may have and don't actually require so you can safely leave them disconnected (e.g. VIP shop if you don't have it)

Many buildings can be off road/grid and function properly, which saves you electricity and fuel that runs the power plants. Any time you start an upgrade on any structure you have to run a road to it to begin, but can immediately take the road back and upgrade will continue and complete properly. It's easier to tell you which structures need constant connection: At least one hospital & powerplant, all houses, radar. tower and depots.
