What's the defense number mean within the defensive line. Do I need to match that number with troops to be able to overtake it minimum ?

This is the bonus "Defense Value" in addition to the total defense facility that you have in your base. Leveling up your Defense line also (and more importantly) increases the total number of defense facility it can hold.
