Evolving is done when your hero reaches certain levels. 10, 30, 70, 130, 210, 310.

To evolve you need a certain amount of combat manuals from the campaign and also some anti serum. In the evolve screen for your hero it will show you what you need for your next evolve. If you are missing Anti serum then you can get more from the virology lab buildings in your sanctuary or from attacking zombie lairs.
