When using diamonds to build and you hit the "build now" button, does it also use resources to build or does it only use the diamonds?

Yes it uses the resources. The diamond cost that you see will be the cost of speeding up the time taken + any missing resources (for example if you don't have enough wood available then it will include the cost of buying the missing wood). Resources needed that you do have available will be consumed with the upgrade.
