First, you have to obtain a relic called Randomazzo . This is found in the northern part of the Gallo Tower stage. It will also be helpful if you have the Milky Way Map relic unlocked, which will point you to the right direction and make this relic visible on the map, making it easier to locate.
This relic is guarded by special enemies that drastically cut your movement speed so it will be very helpful if you have weapons that auto-target enemies (like the Lightning Ring) or at least reach them with ease (any powered-up AoE weapons). After defeating the enemies guarding it, simply pick it up. Like the other relics of the game that unlocks features, you only need to pick up this relic once to permanently unlock the Arcana system.
Alternatively, if you have unlocked the game's built-in Secrets (Cheat Codes) menu, you can just use the cheat code randomazzami to obtain this relic without going through the trouble.
Vampire Survivors: How to Use the Arcana Cards
In the stage selection screen, you can enable Arcanas by ticking the checkbox at the bottom (only visible once you have unlocked this feature). At the start of the stage, you'll be able to pick one of the unlocked cards (or just press the blue question mark icon to randomize your selection). As a general recommendation, pick the arcana which will benefit you immediately. For example, getting an arcana that improves your character's default weapon (and weapons you're planning to collect in the run) will be a good start.
You can only equip up to three Arcana cards at a time. You can obtain more arcana cards by defeating special enemies around the 11-minute mark and the 21-minute mark. These drop purple treasure chests that can contain four random arcana cards. If you get additional cards, you can still choose a new one and discard one that you currently have in exchange.

V - Chaos in the Dark Night
Description: Overall projectile Speed continuously changes between -50% and +50% over 10 seconds. The character starts gaining +1% projectile Speed every level.

IX - Divine Bloodline
Description: Armor also affects listed weapons' damage and reflects enemy damage. Character gains bonus damage depending on missing Health. Defeating enemies with retaliatory damage gives +0.5 Max Health.

X - Beginning
Description: Listed weapons get +1 Amount. The character's main weapon and its evolution gain +3 Amount instead.
