Quests and Challenges are your best bet when it comes to gaining levels. They award lots and lots of experience for tasks that you will be doing anyway. There are instances when you have to use specific raw materials in your board. Rewards are not limited to experience though, some cases quests and challenges reward you with influence points, and gifts.
There can be as many quests as the game wants to throw at you. Basically, quests act as "campaign objectives" for you. You will build the structures or gather the resources needed to establish a territory before moving on to the next one anyway.
Challenges are different beasts. They are randomly generated to the point that it does not make sense. Sometimes the challenges ask you to totally abandon what you are trying to achieve by requiring you to collect resources or materials that you are not focusing on. You can only have two challenges and they will not get replaced until you complete them. Paying runes allow you to get a new pair of challenges. You can also increase the amount of challenges by paying real money.
You can return your previous settlements for more quests. These quests have themes. They usually focus on one aspect of the game that serves as tutorial. For example, there may be a "Carrot Festival" in your first town. This should serve as practice on how to efficiently harvest vegetables for your current town. You can also set-up Royal Quests where you can put up a challenge for you and your friends to compete.
