Is there a set number of flowers required to move up a level, or is it random?

It is not random. It is preset but it goes up and down. The 2nd scene in each chapter requires the least, usually between 60 and 90. The 3rd scene usually requires 170-270. The 4th scene usually requires 270-350. The 5th scene requires 350-580. Moving to a new chapter and opening the 1st scene requires about the same as the 5th scene. In general, expect to need 1200-1900 flowers per chapter.
